Intended Use

L-Type P-AMY is an in vitro assay for the quantitative determination of the amylase isozyme in serum, plasma or urine.

Special Characteristics

L-Type P-AMY uses BG5P as the substrate which is not hydrolyzed by glucoamylase and α-glucosidase. The substrate is stoichiometrically converted to the reaction product. S-amylase activity is inhibited specifically by monoclonal antibody.

Principle of the Method

When anti-S amylase antibody reacts with S-amylase, S-amylase activity is specifically inhibited. A specimen is allowed to react with the BG5P, BG3 and p-nitrophenyl α-maltoside are formed by the action of p-amylase in the specimen. Thus p-nitrophenyl α-maltoside produced is hydrolyzed to p-nitrophenol by the reactions of both glucoamylase and α-glucosidase. By measuring the increase in absorbance due to the production of p-nitrophenol, p-amylase activity in the specimen is determined. A benzyl group in the non-reducing end glucose residue of BG5P prevents the hydrolysis by the coupling enzymes and results in giving low blank. Also, one unit of BG5P hydrolyzed by p-amylase is converted to one p-nitrophenol molecule. Therefore, one unit of α-amylase can be defined stoichiometrically as the amount of enzyme that produces 1 micro mol of p-nitrophenol per minute.
