L-Type CK-MB (2) is a convenient ready-to-use liquid type reagent which utilizes immunoinhibition method with the antibody against CK-M. This reagent using double kinetic mode measurement is not interfered significantly by Myokinase.
Principle of the Method
When a specimen is mixed with Enzyme, containing ADP, hexokinase (HK), glucose-6-phosphate dehydrogenase (G-6-PDH), NADP and anti CK-M antibody, the activity on M-subunit, consisting of CK-MM and CK-MB, is inhibited. By adding Substrate, containing creatine phosphate and glucose, ADP is converted to ATP by the action of remaining B-subunit, in CK-MB. ATP so formed is used to synthesize the glucose-6-phosphate (G-6-P) in a reaction catalyzed by HK. The G-6-P, produced in this way, is oxidized to 6-phosphogluconate in a reaction catalyzed by G-6-PDH, while NADP is reduced to NADPH. The absorbance at 340nm increases along with it. CK-MB activity in the specimen is calculated by the calibrator absorbance change.

Latex Immunoassay
Special Characteristics
L-Type CK-MB mass is designed for general biochemical automated analyzers which is used new enhancer for latex turbidimetric immunoassay. This reagent can avoid the influence of M-subunit and B-subunit. Moreover, it is no significant interference from serum components.
Principle of the Method
When a sample is mixed with Buffer and Latex Reagent, CK-MB in the sample combines specifically with the latex sensitized with anti-human CK-MB antibody (Mouse monoclonal antibody) in the Latex Reagent to yield an insoluble aggregate which causes increased turbidity in the solution. The degree of turbidity of solution can be measured optically and is proportional to the concentration of CK-MB in the patient's sample.
